ABSTRACT

Objective To systematically evaluate the diagnosis value of thyroid autoantibodies in hashimo-to′s thyroiditis. Methods Through the systematic retrieval of the Chinese periodical full text database (CNKI),the full text database of Chinese sci-tech periodicals (VIP),the Wanfang database, the PubMed,EM-base,Scopus,Cochrane library, the database of academic dissertations at home and abroad, the database of the conference papers and so on,all formally published scientific research literature from the establishment of da-tabase to January 2017 were retrieved. The retrieval conditions included "thyroid autoantibody","thyroid per-oxidase antibody"or "TPOAb","thyroid globulin antibody" or "TGAb","thyroid microsome antibody" or "TMAb","thyrotropin receptor antibody"or "TRAb","Hashimoto""Mastitis"or "Hashimoto′s disease".The study that accorded with the standard, the data of thyroid autoantibodies,Hashimoto′s thyroiditis and other data were analyzed by Meta in order to find out the best clinical evidence. Results In the meta-analysis of 33 articles, the levels of thyroid peroxidase antibody (TPOAb),antithyroid globulin antibody (TGAb) and thy-rotropin receptor antibody (TRAb) in the patients with Hashimoto′s thyroiditis were significantly higher than those in the healthy control group. The positive rates of TPOAb,TGAb,TRAb and TMAb in the patients with Hashimoto′s thyroiditis group were higher than those in the healthy control group. The difference was statistically significant (P< 0.05).Conclusion Detection of thyroid autoantibodies TPOAb,TGAb,TRAb and TMAb has important clinical value in the diagnosis of Hashimoto′s thyroiditis.

ABSTRACT

Objective To investigate the effect of levothyroxine(L-T4) intervention on premature delivery occurrence risk in early pregant women complicating subclinical hypothyroidism. Methods The databases of PubMed,EMBASE,EBSCO,Cochrane library,CBM and Wanfangdata were retrieved for collecting the related literatures on pregnancy complicating subclinical hypothyroidism published from January 1980 to March 2017.The undesirable studies were deleted according to the inclusion standard. The meta analysis was performed by using the RevMan5.3 software. Results (1) The 12 articles on premature delivery occurrence risk in pregnan women complicating subclinical hypothyroidism without intervention were included(accumulated 5 612 cases of samples and accumulated 368 cases of premature delivery),802 pregnant women received L-T4 intervention, the control group (4 810 cases) was the pregnant women of normal thyroid. The merged OR was 3.46,95%CI 2.64-4.54,P<0.05.(2) Twenty-one articles on premature delivery occurrence risk in pregnan women complicating subclinical hypothyroidism treated by L-T4 in meta analysis were included(accumulated 2561 cases of samples and accumulated 286 cases of premature delivery). The merged ORwas 5.37,95%CI 3.90-7.38,P<0.05).Conclusion Early pregnancy complicating subclinical hypothyroidism can increase the risk of preterm birth, whereas adopting the L-T4 intervention therapy can significantly reduce the risk of premature delivery occurrence.

ABSTRACT

BACKGROUND: Chinese medicine Gukang prescription has a clear effect on clinical treatment of osteoporosis, but the therapeutic pathway is stil unclear. OBJECTIVE:To investigate the effects of Chinese medicine Gukang on the expression of receptor activator of nuclear factor kappa B ligand and osteoprotegerin by regulating core binding factor alpha 1 expression to control the growth and development of osteoblasts. METHODS:Sprague-Dawley neonatal rats within 24 hours after delivery were used for the separation and culture of osteoblasts. Adult Sprague-Dawley rats were used to prepare drug-containing serum, and then divided into two groups randomly:normal control group and Gukang group. Rats in the normal control and Gukang groups were intragastrical y administrated with extract of Gukang prescription and normal saline based on rat’s body surface area, for 1 consecutive week. Two hours after the last administration, blood samples were taken from the heart. Then the serum was col ected. Osteoblasts at passage 3 were confirmed with alkaline phosphatase assay and digested. After counting and planting, al osteoblasts were divided into two groups and treated with col ected serum for 72 hours. Proliferative rate of osteoblasts was detected by using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide test. Secretion of alkaline phosphatase was detected by using enzyme linked immunosorbent assay and corrected with the corresponding absorbance value. mRNA expression of core binding factor alpha 1, receptor activator of nuclear factor kappa B ligand and osteoprotegerin were detected by using reverse transcription-PCR in al groups. RESULTS AND CONCLUSION:mRNA expression of osteoprotegerin and core binding factor alpha 1 in the Gukang group was significantly higher than that in the normal control group, but protein and mRNA expression of receptor activator of nuclear factor kappa B ligand were dramatical y lower in the Gukang group compared with the normal control group (Pcore binding factor alpha 1, thereby adjusting the expression of receptor activator of nuclear factor kappa B ligand and osteoprotegerin, which may be one of the mechanisms underlying Gukang treatment for osteoporosis.
